Nowadays, cancer has been the main cause of death. One of the characteristic of the cancer cells is its low adhesive force among cells which means that it's easier for them to fall into the blood circulation system and become circulating tumor cell, CTC. The detection of the type and number of CTC is meaningful to the treatment of cancer. However, CTCs are extremely rare compared with blood cells, ranging from 1 to 100 in 1ml blood. Therefore, effective isolation and enumeration of CTCs have been a great technical challenge. In our experiment, we design a system based on microfluidic chip to achieve CTC filtration and enumeration. The technology mainly takes advantage of nano-imprinting , thermoplastics forming and photoetching. Polydimethylsiloxane (PDMS) is used as the basic material for the device. To promote the filtration efficiency and the recovery efficiency, a circulating filtration system has been devised. This project developed an integrated CTCs isolation and enumeration lab-on-chip system , which utilized microfluidic devices as the central processing unit. The filtration and enumeration device was mainly fabricated by PDMS soft lithography and transfer-bonding. In initial testing, the filtration efficiency is able to reach 70% and the recovery efficiency can reach 90%. The fluorescence signal of stained CTC cells could be detected by a fluorescence microscope. The structure of this CTC analysis system is simple. It is not only convenient to operate, but also is low-cost, thus holds the potential to be developed into a practical CTC detection tool for early cancer diagnosis
